Mutation that leads to the permanent production of a (defective) gene product that can no longer be regulated. Causes of this are, for example, mutations in a regulator gene whose product functions as a repressor, or a mutation in the sequence of a repressor-binding cis-element. However, changes in a promoter gene are also possible.
