CYBB (CYBB steht für „Cytochrom B-245 Beta-Kette“) ist ein Protein kodierendes Gen das auf Chromosom Xp21.1-p11 lokalisiert ist. Zu den assoziierten Signalwegen gehören die Klasse I MHC-vermittelte Antigenverarbeitung und -präsentation und die G-Protein-Signalübertragung RAC1 in zellulären Prozessen.

Allgemeine Information

Cytochrom b (-245) setzt sich aus der Alpha- (CYBA) und der Beta-Kette (CYBB) zusammen. Es fungiert als Hauptbestandteil des mikrobiziden Oxidase-Systems der Phagozyten.

Ein CYBB-Mangel ist einer der fünf beschriebenen biochemischen Defekte, die mit der chronischen granulomatösen Erkrankung (CGD) in Verbindung gebracht werden. Bei dieser Erkrankung ist die Aktivität der phagozytären NADPH-Oxidase vermindert; die Neutrophilen sind in der Lage, Bakterien zu phagozytieren, können sie aber nicht in den phagozytären Vakuolen abtöten. Die Ursache für den Tötungsdefekt ist die Unfähigkeit, die Zellatmung zu erhöhen, und die daraus folgende Unfähigkeit, aktivierten Sauerstoff in die phagozytische Vakuole zu liefern.

Das kodierte Protein ist eine wichtige kritische Komponente der membrangebundenen Oxidase von Phagozyten , die Superoxid erzeugen. Das Protein ist die finale Komponente einer Atmungskette, die einzelne Elektronen vom zytoplasmatischen NADPH durch die Plasmamembran auf molekularen Sauerstoff an der Außenseite überträgt. Es fungiert auch als spannungsgesteuerter Protonenkanal, der die H(+)-Ströme ruhender Phagozyten vermittelt und ist an der Regulierung des zellulären pH-Wertes beteiligt. Das Protein wird durch Zink blockiert.
